Abstract
Inositol polyphosphate multikinase (IPMK) is a versatile enzyme with dual inositol and lipid kinase activities, crucial for generating key signaling molecules like InsP5 (inositol pentaphosphate), InsP6 (inositol hexaphosphate), and PIP3 (phosphoinositide 3,4,5 tri phosphate). Originally identified in yeast and later found essential for mammalian embryonic development, IPMK regulates diverse nuclear and cytoplasmic pathways, including nutrient sensing, growth factor signaling, mRNA transport, and transcription. Two decades of multidisciplinary research have unveiled IPMK's complex biological functions and its potential role in neurodegenerative diseases, cancer, inflammation, and other diseases. This review examines recent IPMK research, focusing on its structure-function relationships, diverse cellular roles, and physiological impacts.
